非洲猪瘟TaqMan实时荧光定量PCR检测方法的建立与应用

为建立检测非洲猪瘟病毒(African swine fever virus,ASFV)的快速诊断方法,本研究根据ASFV SY18株p17蛋白的编码基因D117L的保守序列设计并合成引物和探针,建立了基于ASFV p17蛋白的编码基因D117L的TaqMan荧光定量PCR检测方法,并验证其特异性、灵敏性和重复性。结果显示,本研究建立的TaqMan荧光定量PCR检测方法的C_t值与标准品在1×10^9~1×10^1copies/μL范围内呈良好的线性关系,相关系数为0.998,斜率为-3.192,检测下限为10 copies/μL,且与其他能引起相似症状的猪源病毒无交叉反应。重复性试验结果显示,组间与组内变异系数均小于1.920 7%,重复性好。此方法可用于ASF的早期诊断和ASFV快速检测。     

基于钟基因Clock和Bmal1调控探究辰时针刺自发性高血压大鼠的降压机制

目的 以自发性高血压大鼠（spontaneous hypertensive rat，SHR）为观察对象，探究针刺降压的钟基因调控作用。方法 将24只SHR随机分为针刺组和模型组，12只Wistar-Kyoto（WKY）大鼠作为正常组。针刺组选择辰时针刺SHR双侧曲池、足三里穴，模型组和正常组仅接受与针刺组同等强度的捆绑操作。4周后，各组分别在辰、酉时随机抽取6只大鼠，测量鼠尾收缩压（systolic blood pressure, SBP）和舒张压（diastolic blood pressure, DBP），然后检测血清5-羟色胺（5-hydroxytryptamine，5-HT）、褪黑素（melatonin, MT）的含量以及心脏生物钟基因Clock和Bmal1的表达水平。结果 分组因素对SBP、DBP、血清MT含量、Clock表达结果的RQ值、Bmal1表达结果的RQ值的主效应均有统计学意义（P＜0.05），时辰因素对各指标的主效应和两者的交互作用均无统计学意义（P＞0.05）。模型组辰、酉两个时辰的SBP和DBP均高于正常组（P<0.05），针刺组辰时的SBP和DBP及酉时的SBP均低于模型组（P<0.05）。模型组辰时MT含量较正常组降低，5-HT含量较正常组升高（P<0.05），针刺组辰时MT含量较模型组升高，5-HT含量较模型组降低（P<0.05）；针刺组酉时的MT含量较模型组升高（P<0.05）。针刺组辰时心脏Clock基因表达水平和酉时心脏Bmal1基因表达水平较模型组升高（P＜0．05）。结论 辰时针刺可以降低SHR的“双峰”血压，其作用机制可能与提高血清MT含量，减少5-HT含量以及提高心脏Clock和Bmal1基因的表达水平有关。     

复发性流产肾虚证小鼠子宫蜕膜组织miRNA表达谱检测及生物信息学分析

目的 探讨微小RNA（microRNA，miRNA）在复发性流产（repeated spontaneous abortion，RSA）肾虚证小鼠子宫蜕膜组织中的表达特征。方法 运用基因芯片技术对所采集的正常对照组和RSA小鼠蜕膜组织进行miRNA检测，运用Genespring软件对miRNA进行筛选及功能分析。应用生物信息学方法分析差异表达的miRNA及其靶向基因在RSA发生、发展中的作用。结果 芯片检测结果显示正常对照组和RSA肾虚证组筛选出差异表达的miRNA共有63条（22条上调、41条下调），实时荧光定量PCR验证的miRNA的表达情况与miRNA芯片结果一致。京都基因与基因组百科全书通路富集分析及基因本体分析显示，其差异表达的miRNA涉及到PI3K-Akt信号通路、细胞吞噬通路、癌症通路、黏着斑通路、肌动蛋白细胞骨架调控通路、轴突导向信号、MAPK信号通路、PAP-1信号通路等。结论 RSA肾虚证小鼠子宫蜕膜组织中存在差异表达的micro RNA，可能涉及RSA肾虚证的发生、发展。     

Brandom on Norms and Objectivity

The aim of this paper is to investigate Brandom’s conception of the objectivity of norms. In Making It Explicit Brandom supports a weak notion of objectivity based on his understanding of the perspectival structure of linguistic practices. In his following works, he resorts to the Hegelian notion of recognition, adding a historical dimension to his account. I contend that this notion of objectivity can be successfully defended against the objections raised by the commentators. In particular, it does not jeopardise the same possibility of communication, as claimed by Habermas and others, unless a strongly objective notion of communication is assumed. However, the paradigm shift from a strong to a weak understanding of objectivity entails a consequent revision of the conception of social criticism.     

基于ITS2条形码序列对新型“香料”毒品植物成分的研究

目的应用DNA条形码片段ITS2对新型毒品样品中的植物成分进行分析。方法提取可疑毒品样本"spike99","K2","7号"中的植物基因组DNA,用ITS2通用引物进行扩增,对扩增产物进行测序,对序列校对拼接,应用BLAST方法在NCBI数据库中比对。结果 "spike 99"、"7号"各得到1条ITS2基因序列,"K2"得到2条ITS2基因序列,分别与紫花苜蓿、大麻、啤酒花、黄蜀葵的ITS2基因序列的同源性为100%。结论应用条形码技术可以成功分析新型毒品中的植物种属,为案件中植物样本的种属鉴定提供方法。     

I don't Agree with Nick: Retrodicting the Conservative–Liberal Democrat Coalition

Insider and journalistic accounts of the formation in May 2010 of the Conservative‐Liberal Democrat coalition actually, although not explicitly, rely on types of explanation familiar to those who study politics. They tell us that structure (or at least the economy) was important. So too, they suggest, were institutions (timing and the rules of the game). They also stress the importance of contingency (‘events, dear boy, events’) and agency (who did and said what to whom). While none of these things were unimportant, they only served to make certain an outcome that anyone with a passing acquaintance with the theory and the practice of coalition formation would have predicted—namely a ‘minimum winning coalition’. The only thing that could have made that outcome uncertain was a fundamental ideological difference between the two parties involved; however, it quickly became apparent—to the surprise of those of us who failed to appreciate how much the Liberal Democrats had changed—that no such difference existed. Indeed, it is possible to argue that the coalition formed was not merely minimum winning but ‘minimum connected winning’. As such, its formation was not so much breathtakingly bold and exciting as pretty much inevitable. In the end, the maths and the physics mattered more than the chemistry. Fortunately for the Conservatives, the Liberal Democrats seriously underplayed their hand in the negotiations, with possibly disastrous consequences for them in the long term.

小布什入主白宫后，美国新政府便开始调整对华战略，将克林顿时期的对华“接触　＋遏制”战略，改为“遏制＋接触”。其原因是小布什政府看到了对华战略的侧重点发生了　重大变化，其动机是企图达到谋取建立世界政治、军事、经济和文化霸权的目的。美国调　整对华战略，必将对我国的国家安全造成新的威胁，对此，我们应予以正确的评估。     

浅谈公安现役院校“红色基因”传承

新形势下,世情、党情、国情发生深刻变化,意识形态领域斗争尖锐复杂,公安现役院校只有不断挖掘"红色基因"这一优势资源,将其引入院校人才培育过程中来,通过加强顶层设计、丰富培育形式、壮大建设人才队伍来提升传承"红色基因"的时效,才能充分发挥好政治工作生命线的地位作用,才能真正做到"把红色基因融入官兵血脉,让红色基因代代相传"。     

广东省猪源葡萄球菌耐药性分析及多重耐药基因cfr的流行现状

为了解2013-2017年间广东省猪源葡萄球菌的耐药情况及多重耐药基因cfr的流行现状,采集生猪养殖场和交易市场猪鼻腔拭子分离葡萄球菌,采用琼脂稀释法测定分离菌株对14种抗菌药物的敏感性,并用PCR检测mecA和cfr基因的携带情况。结果显示,在采集的4099份样品中分离到葡萄球菌1485株,其中金黄色葡萄球菌173株。猪源葡萄球菌对红霉素、四环素、泰妙菌素、克林霉素、沃尼妙林和氟苯尼考耐药严重,耐药率分别为91.0%、90.2%、87.7%、84.6%、84.6%及82.0%,对利福平和利奈唑胺相对敏感,未发现万古霉素耐药菌株;庆大霉素、四环素、红霉素和环丙沙星的耐药率呈现逐年上升的趋势。在1485株葡萄球菌中,mecA和cfr的检出率分别为39.9%和12.3%。cfr阳性菌株常携带多种耐药基因,cfr基因的遗传背景也呈现多样化。结果表明,广东省猪源葡萄球菌的耐药情况严重,cfr的检出率高,应加强养殖业中抗菌药物的规范和合理使用,减缓耐药性的产生和传播。     

PowerPlex~ 21试剂盒扩增后的分型图谱中Y片段丢失的识别

目的依据分型图谱,直观、简便识别是否存在Amelogenin基因Y片段丢失的可能。方法采用计算分型图谱中等位基因峰高之和比值的方法,统计1 968份人员样本经Power Plex~ 21试剂盒扩增后Amelogenin与D3S1358基因座之间的均衡性、Amelogenin X等位基因与Amelogenin Y等位基因的均衡性以及D3S1358基因座不同等位基因的均衡性情况。结果 90.8%的女性样本Amelogenin X等位基因峰高不低于D3S1358基因座等位基因峰高和的60%,94.9%的男性样本Amelogenin X等位基因的峰高不超过D3S1358基因座等位基因峰高和的70%。结论 Power Plex~ 21试剂盒扩增后的分型结果中,当Amelogenin基因只检测到Amelogenin X等位基因,且Amelogenin X等位基因的峰高不及D3S1358基因座等位基因峰高之和的70%时,应考虑存在Y片段丢失的可能性。
